In viltro Antibacterial Activity of WCM 302, a Substance Obstained from Culture



Abstract
Over 600 isolates of Streptomyces sp. from soil were screened to find strains producing antibiotics active against resistant Staphylococcus aureus and Streptomyces sp. 302 was selected. From the culture filtrate of the surain. the active compound
was
purified and the activity against various microorganisms including 150 isolates of S, aureus was determined by an agar dilution method.
Minimal inhibitory concentrations (MICs) of WCM 302 were 0.03 §¶/ml for Gram positive bacteria, 0.47§¶/ml-7.5§¶/ml for E. coli, 7.5§¶/ml for S. typhimurium and higher than 30§¶/ml for other Gram-negative bacteria or fungi. This compound was found
to be
active against 150 strains of drug-resistant S. aureus isolated in hospital. MICs for 150 clinical isolates of drugresistant S. aureus were 0.004§¶/ml-0.9§¶/ml and 90% MIC were less than 0.2§¶/ml. Multi-drug resistant and highly resistant S.
aureus
were
also inhibited at the concentrations of 0.004§¶/ml-0.9§¶/ml. Antibacterial activity of the compound was 10 times more active than vancomycin against S. aureus and there was no association between antibacterial activity of this compound and
vancomycin.
In conclusion, WCM 302 purified from the culture of Streptomyces sp. 302 was active against Gram-positive bacteria including 150 clinical isolates of S. aureus. The different pattern of activity of the coumpound and those of 14 antibiotics
including
vancomycin against the S. aureas isolates indicate that the compound may be a novel material not related to the antibiotics tested.
